杠板归总黄酮对小鼠酒精性肝损伤的保护作用

摘要:

【目的】探究杠板归总黄酮的保肝药作用及其作用机理,为杠板归的深入研究提供活性物质基础和理论依据。【方法】随机将60只小鼠分为正常组、模型组、联苯双酯阳性组(150 mg/kg)及杠板归黄酮高剂量组(600 mg/kg)、中剂量组(400 mg/kg)、低剂量组(200 mg/kg),除正常组外,其余各组按16 mL/kg剂量灌胃56%vol白酒,1 h后各用药组按20 mL/kg剂量灌胃给药,正常组和模型组灌胃等体积蒸馏水,连续给药14 d,然后通过计算肝脏指数(LI)、脾脏指数(SI)、胸腺指数(TI),生化测定血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、丙二醛(MDA)和超氧化物歧化酶(SOD),ELISA检测肝组织中白细胞介素-4(IL-4)、白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)等,探究杠板归总黄酮对小鼠酒精性肝损伤的保护作用。【结果】各杠板归总黄酮剂量组可有效降低因酒精性肝损伤引起小鼠升高的LI,并升高SI和TI;又能降低血清ALT、AST活性及MDA含量,升高SOD活性;还可降低酒精性损伤肝组织中的IL-4、IL-6和TNF-α含量,且对小鼠肝小叶结构、肝细胞坏死和变性及炎细胞浸润等方面均有改善作用,尤其以杠板归总黄酮高、中剂量组的效果最明显。【结论】杠板归总黄酮对小鼠急性酒精性肝损伤具有保护作用,其机理是通过抗脂质过氧化、抑制肝脏炎症反应来实现保肝作用。

【Objective】In order to provide material and theoretical basis for further studying Polygonum knotweed L. on active compounds in the future, the present experiment was conducted to study hepatoprotective effect and mechanism of total flavonoids extracted from P. knotweed . 【Method】Sixty mice were randomly divided into 6 groups viz., normal group, model group, bifendate positive group(150 mg/kg), high-dose total flavonoids from P. knotweed group(600 mg/kg), medium-dose total flavonoids group(400 mg/kg) and low-dose total flavonoids group(200 mg/kg). These groups were intragastrically administrated with 56%vol alcohol at a dose of 16 mL/kg per day, except normal group, and after 1 h, which were intragastrically administrated with total flavonoids at a dose of 20 mL/kg per day, and the normal group and model group were given isopyknic of distilled water every day for 14 d. Then the liver index(LI), spleen index(SI) and thymus index(TI) were calculated, and the alanine aminotransferase(ALT), aspartate transaminase(AST), malondialdehyde(MDA) and superoxide dismutase(SOD) in serum were determined by biochemical method. The interleukin-4(IL-4), interleukin-6(IL-6) and tumor necrosis factor-α(TNF-α) in liver tissue were detected by ELISA. Based on above results, the protective effect of total flavonoids from P. knotweed on mice with alcoholic liver injury was investigated. 【Result】In all total flavonoids groups, the LI of mice with alcoholic liver injury was obviously reduced, the SI and TI were increased, the activities of ALT and AST and content of MDA in serum were reduced, and the activity of SOD was increased in mice with alcoholic liver injury. Furthermore, total flavonoids from P. knotweed could also reduce content of IL-4, IL-6 and TNF-α in the liver tissue of mice with alcoholic injury, and improve hepatic lobules structure, hepatic cell necrosis and degeneration, and inflammatory cell infiltration, especially high-dose and medium-dose total flavonoids. 【Conclusion】The total flavonoids from P. knotweed have protective effect on mice with acute alcoholic liver injury, their functional mechanism may be related to anti-lipid peroxidation and inhibition of liver inflammation.
