pH胁迫对脊尾白虾代谢酶活力的影响 |

【目的】研究pH胁迫对脊尾白虾(Exopalaemon carinicauda Holthuis)代谢酶活力的影响,丰富脊尾白虾的逆境生物学和代谢生物学基础理论,同时为其养殖环境的调控提供科学指导。【方法】设置5.5、7.0、8.5和10.0 4个pH梯度,分别于胁迫0、1、3、6、12、24和36 h时从脊尾白虾心脏抽取血淋巴,分离血清,然后检测酸性磷酸酶(ACP)和碱性磷酸酶(AKP)活力变化。【结果】短期(3 h)pH胁迫下,偏酸性环境可提高脊尾白虾血清ACP活力,而偏碱性环境会刺激脊尾白虾血清ACP活力下降。pH 8.5和pH 10.0胁迫组脊尾白虾血清ACP活力随胁迫时间的延长整体上呈降低—升高—降低的波浪式变化趋势,而pH 5.5和pH 7.0胁迫组恰好相反,整体上呈升高—降低—升高的变化趋势。在AKP活力方面,pH胁迫后脊尾白虾血清AKP活力迅速提高,整体上表现为:pH 7.0和pH 8.5胁迫组脊尾白虾血清ACP活力随胁迫时间的延长呈先升高后降低的变化趋势,pH 5.5和pH 10.0胁迫组则呈逐渐缓慢升高的变化趋势。【结论】在pH 7.0~8.5范围内,脊尾白虾的磷酸酶可迅速作出反应,促使机体自我调整至相对稳定状态,但过酸或过碱的养殖水体会显著影响其非特异性免疫力。 |
Objective】The effects of pH stress on the activities of phosphatase in Exopalaemon carinicauda Holthuis were studied to rich the basic data for stress and metabolism biology and supply the theoretical direction for aquaculture environment regulation. 【Method】Four pH values(5.5, 7.0, 8.5, 10.0) were designed. The activities of acid phosphatase (ACP) and alkaline phosphatase(AKP) in serum of E. carinicauda Holthuis were detected at 0, 1, 3, 6, 12, 24, 36 hours after different pH stress. 【Result】The results showed that short time pH stress within three hours, ACP activities increased in an acid environment and decreased in an alkaline environment. ACP activities presented “decrease-increase-decrease” wavy change trend at pH 8.5 and 10.0 treatments, but the opposite change trend presented at pH 5.5 and 7.0 treatments. AKP activities increased rapidly after pH stress, presented “increase-decrease” change trend at pH 8.5 and 10.0 treatments, and increased slowly at pH 5.5 and 10.0 treatments in the course of the experiment. 【Conclusion】Phosphorylase can respond quickly and adjust to relative stable state in pH 7.0-8.5, but redundant acid or alkali can affect the non-specific immunity of .E. carinicauda Holthuis. |
