基于SSR分子标记的广西柿种质资源遗传多样性分析

【目的】采用SSR标记技术分析广西柿种质资源的遗传多样性,为其资源的分类、鉴定、保护和开发利用提供依据。【方法】设计48对SSR引物,对从广西不同生态气候区收集的93份柿种质资源进行PCR扩增、聚类分析和主成分分析,探讨不同种质资源的遗传多样性和亲缘关系。【结果】在48对引物中,15对SSR引物可从93份柿种质中扩增获得159条带,多态性条带比例为96.86%;各种质间的遗传相似系数在0.478~0.925,平均为0.667。聚类分析结果显示,在遗传相似系数0.640处可将93份柿种质资源分为A、B、C、D 4组,其中柿栽培种、油柿和君迁子分属不同组中,野柿与柿栽培种间存在交叉现象。主成分分析结果表明,前三主成分贡献率依次为14.18%、5.25%和4.11%,累积贡献率为23.54%;主成分分析可将93份柿种质材料分为四大类,与聚类分析结果基本一致。【结论】广西柿种质资源遗传背景广、多样性较丰富,可为柿资源开发利用和新品种选育提供重要材料;SSR分子标记可作为柿种质资源分类和鉴定的可靠工具。

【Objective】The present study was conducted to analyze the genetic diversity of Diospyros germplasm resources in Guangxi by using SSR molecular marker in order to provide references for classification, identification, protection and utilization of Diospyros resource. 【Method】The 93 accessions of Diospyros germplasms collected from different eco-climatic regions of Guangxi were amplified by using 48 pairs of SSR primers, and then the cluster analysis and principal component analysis (PCA) were conducted to detect their genetic diversities and relationships. 【Result】Out of 48 pairs of primers, 15 pairs of primers could amplify 159 bands, and 154 of which were polymorphic loci with a polymorphism rate of 96.86%. The similarity coefficient between different germplasms varied from 0.478 to 0.925, with an average of 0.667. The cluster analysis conducted by unweighted pair-group method with arithmetic means (UPGMA) showed that, 93 accessions of Diospyros germplasms could be clusted into 4 groups at similarity coefficient of 0.640. The germplasms which belonged to Diospyros kaki Thunb, D. oleifera Cheng and D. lotus Linn were clustered into different groups. An overlapping phenomenon appeared in the genetic relationship between D. kaki var. silverstris Mak and Diospyros kaki Thunb. The results of PCA showed that, the contribution rate of the first three principal components were 14.18%, 5.25% and 4.11% in sequence, and the accumulated contribution rate was 23.54%. The PCA also could cluster 93 accessions of germplasms into 4 groups, which was similar to that of cluster analysis. 【Conclusion】The Diospyros germplasm resources in Guangxi present wide genetic background and abundant diversity, which can be used as important materials for utilization and breeding of Diospyros germplasm resources. SSR molecular marker can be used as a reliable tool for classification and identification of Diospyros germplasm resources.
