【Objective】The experiment was conducted to analyze the genetic variation amongst different mulberry diploids and their homologous tetraploids to understand the genetic mechanism of multagenesis in mulberry polyploids. 【Method】The polymorphism amongst diploid and homologous tetraploid of 8 mulberry varieties was assessed using RAPD molecular marker technique and the genetic diversity was analyzed using cluster analysis. 【Results】The results showed that RAPD polymorphism between diploid and their homological tetraploid varied significantly in different mulberry varieties. Differential bands were not observed amongst diploid and their homological tetraploid in Nongsang 8 and Husang 199 varieties, while the highest RAPD polymorphism (20.41%) between diploid and its homological tetraploid was observed in Husang 32, followed by Tongxiangqing, Huangsang 37, Husang 197, Guosang 20 and Yu 71-1,which was 9.09, 6.06, 5.45, 5.17 and 3.17%,respectively. The cluster analysis results clearly indicated the presence of two distinct clusters of the mulberry varieties used in the assessment. Husang 32 and Tongxiangqing were found in the same cluster. 【Conclusion】The colchicine treatment can cause a significant genetic variations in different diploids of mulberry genotypes.
